qScript™ One-Step Fast qRT-PCR Kit

Rapid One-Step qRT-PCR with Broad Dynamic Range and High Sensitivity (ACTB)
Beta actin mRNA was amplified from log-fold serial dilutions of HeLa cell total RNA (1 µg to 1 pg) in 10-µL volumes with the qScript™ One-Step Fast qRT-PCR Kit and FAM-BHQ1 labeled 5’-hydrolysis probe. Triplicate reactions for each RNA quantity were incubated for 5 min at 50ºC followed by 30s at 95ºC and then 40 cycles of 3s, 95ºC; 20s, 60ºC on a Mastercycler® ep realplex S (Eppendorf). Total run time was under 41 minutes.

Glyceraldehyde-3-phosphate dehydrogenase mRNA was amplified from log-fold serial dilutions of HeLa cell total RNA (1 µg to 1 pg) in 10-µL volumes with the qScript™ One-Step Fast MGB qRT-PCR Kit and GAPD (FAM-MGB) TaqMan® Endogenous Control primer/probe set (Applied Biosystems). Triplicate reactions for each RNA quantity were incubated for 5 min at 50ºC followed by 30s at 95ºC and then 40 cycles of 3s, 95ºC; 20s, 60ºC on a Mastercycler® ep realplex S (Eppendorf). GAPD mRNA was amplified from log-fold serial dilutions of HeLa cell total RNA (1 ug to 1 pg) in 25-uL volumes with the qScript™ One-Step qRT-PCR Kit and GAPD TaqMan® Endogenous Control primer/probe set (Applied Biosystems) according to the manufacturers protocol. Averaged plots of quadruplicate qRT-PCR reactions are shown.
